An “Omic” Overview of Fragile X Syndrome

Fragile X syndrome (FXS) is a neurodevelopmental disorder associated with wide range of cognitive, behavioral and medical problems. It arises from the silencing fragile mental retardation 1 (FMR1) gene and, consequently, in absence its encoded protein, FMRP (fragile protein). ubiquitously expressed multifunctional RNA-binding primarily considered as translational regulator. Pre-clinical studies past two decades have therefore focused on this function to relate FMRP’s molecular mechanisms underlying FXS physiopathology. Based these data, successful pharmacological strategies were developed rescue phenotype animal models. Unfortunately, results did not translate into humans clinical trials using same therapeutic approaches reach expected outcomes. These failures highlight need put perspective different functions order get more comprehensive understanding pathophysiology. This work presents review involvement noteworthy that may ultimately contribute various biochemical alterations composing phenotype.
